The Laboratory of Immunology of the Heart Institute/Division of Clinical /Immunology and Allergy is a multidisciplinary lab where facilities for advanced techniques in cellular and molecular immunology, immunogenetics, protein biochemistry and immunochemistry, including proteomics/peptide mass fingerprinting, multiple peptide synthesis, and cDNA microarray immunology are used intensively across the several projects.
The Laboratory has had a long standing interest in the identification of tissue antigens recognized by antibodies and T cells in the context of transplant rejection and post-infectious organ-specific autoimmune diseases, like Chagas disease cardiomyopathy, rheumatic heart disease, Harada’s uveitis.
More recently, with the advent of high-throughput tecnlologies, identification of gene and protein expression profiles from the tissue sources are yielding information on disease-induced changes, that can be validated with the partner technology.Currently, we are using proteome technology for the following projects:
Identification of protein expression differences in heart tissue samples from patients with different etiologies of heart disease. Prelimilary results indicate substantial diffferences in expression of enzymes involved in energy metabolism (lowe in Chagas disease)Identification of proteins expressed in Brazilian wasp venom that are recognized by IgE antibodies from serum of patients allergic to wasp venomIdentification of protein expression differences in heart valve samples from rheumatic heart disease patients or normal donors.
